The Organization of Mouse and Human Cortico-Hippocampal Networks Estimated by Intrinsic Functional Connectivity

Abstract: While the hippocampal memory system has been relatively conserved across mammals, the cerebral cortex has undergone massive expansion. A central question in brain evolution is how cortical development affected the nature of cortical inputs to the hippocampus. To address this question, we compared cortico-hippocampal connectivity using intrinsic functional connectivity MRI (fcMRI) in awake mice and humans. We found that fcMRI recapitulates anatomical connectivity, demonstrating sensory mapping within the mouse … Show more

“…The aCC contains axons related to the somatomotor system connecting the two hemispheres, and the integrity of this system is reflected in rotarod performance ( 41 ). As expected, the control group showed homotopicity (symmetric bilateral correlations), a hallmark of rs-fMRI in humans and rodents ( 29 , 32 , 34 , 35 , 42 ). In contrast, Plp- Nf1 fl/+ mice showed reduced interhemispheric connectivity ( Fig.…”

Exaggerated anticipatory anxiety is common in social anxiety disorder (SAD). Neuroimaging studies have revealed altered neural activity in response to social stimuli in SAD, but fewer studies have examined neural activity during anticipation of feared social stimuli in SAD. The current study examined the time course and magnitude of activity in threat processing brain regions during speech anticipation in socially anxious individuals and healthy controls (HC). Method Participants (SAD n = 58; HC n = 16) underwent functional magnetic resonance imaging (fMRI) during which they completed a 90s control anticipation task and 90s speech anticipation task.

“…Since such algorithms have yet to be adapted to mice, and are beyond the scope of the current work, we used anatomical parcellation based on the Allen Mouse Brain Atlas, which is comprised of 86 cortical labels 33,34 . Previous studies in mice showed close agreement between structural and functional connectivity in the mouse brain 27,54 , including in gross cortical organization 55 . Moreover, using the Allen Mouse Brain Atlas, we have recently shown that structural connectomes can predict their functional counterpart 30 , suggesting that this anatomical parcellation is functionally relevant.…”

“…This could be due to the difference in how reproducibility was measured in the task and resting fMRI (see more discussion in the reproducibility subsection below). Furthermore, the use of GSR led to reduced or even negative FC similar to what has been reported in rodents (Bergmann et al, 2016;Kalthoff et al, 2013) and humans (Murphy et al, 2009). Therefore, the results discourage the use of GSR in rodent rsfMRI preprocessing.…”
